Marc Lubberstedt to Speak on 3D Liver Cell Cultures at Tissue Models Conf., May 6-7, 2014, Berlin

    MONROVIA, CA, January 30, 2014 /24-7PressRelease/ -- Marc Lübberstedt will present on "Bioreactor Studies of Drug Metabolism and Metabolic Functions in Perfused 3D Liver Cell Cultures" at the Tissue Models & Drug Screening Conference, to be held May 6-7, 2014 in Berlin.

Lübberstedt is a member of the Bioreactor Group at the Brandenburg Center for Regenerative Therapies at Charite Universitätsmedizin Berlin. The cultivation of primary human hepatocytes provides a suitable model for hepatic drug metabolism studies. Lübberstedt will present about a scalable 3D-bioreactor for the maintenance of cells in a 3D arrangement, which is composed of hollow fibers for the perfusion with culture medium and decentralized oxygenation of the cell compartment via different fiber layers. The bioreactor provides improved culture conditions for high-density liver cell cultures and can support the reorganization of parenchymal and non-parenchymal cells to tissue-like structures.

The investigations of metabolic functions and drug metabolism of primary human hepatocytes cultured in the 3D-bioreactor shows prolonged hepatocyte-specific functions such as urea production and the stabilization of human-relevant cytochrome P450 in terms of activity and gene expression under serum-free conditions, compared to conventional 2D cultures. Bioreactor cultures of the HepaRG cell line were investigated as an alternative to primary human hepatocytes, and results showed stable CYP activities over a period of two to four weeks. Thus, the 3D hollow fiber perfused bioreactor for high-density liver cell culture could provide a suitable model for toxicity studies and the investigation of drug metabolism on human hepatic cells.
